This session aimed to run a manuscript specialist tool on the EAC (Eosinophilic Esophagitis) program to analyze and improve the quality and scientific rigor of an associated manuscript.

The agent iteratively developed a manuscript (progressing through manuscript_v1–v3.md drafts, an outline, and supplementary information files) alongside a two-round peer review process, producing review reports and point-by-point responses to reviewers for each round (response_to_reviewers_round1.md and round2.md). Five figures (Fig 1–4 and Fig S1, including a GTEx-based selectivity figure) were generated and embedded into the final manuscript. Corrections arising from review were consolidated across all deliverables, notably a selectivity-recalibration finding that GUCY2C's rank-1 target status is contingent on an optimistic selectivity score, with CDH17 retained as a backup target; the final corrected versions were then converted into polished Word documents.

The primary deliverables are four Word documents built from the corrected final versions: manuscript_and_supplements.docx (full manuscript and supplementary information with all five figures embedded), peer_review_report.docx (both review rounds, editor letters, responses, and a revision-tracking table), executive_summary.docx, and lay_abstract.docx. Supporting data remain in separate supplementary CSV files (S1_binder_designs.csv, S2_target_nomination.csv, S3_competitive_landscape.csv, S4_gtex_selectivity.csv, S5_binder_liabilities.csv), which were not folded into the manuscript DOCX as of this session's end.
